 Scenario 1:

 $ killall -9 <pulseaudio PID>
 $ arecord -D hw:0 -f cd test.wav
 $ # (blah, blah, blah...) into the microphone
 $ aplay -D hw:0 test.wav
 $ # (blah, blah, blah...) heard on the speakers

 Scenario 2:
 $ pulseaudio
